Biological Background: TAK1 Function and Localization
- TAK1 (MAP3K7) is a mitogen-activated protein kinase kinase kinase that serves as an essential component of the MAP kinase signal transduction pathway, mediating cellular responses to environmental changes. Related references: PMID:10094049 | PMID:11460167 | PMID:12589052
- It transduces signals from diverse stimuli including TRAF6, IL-1, TGF-beta, TLR, CD40, and BCR by acting as an upstream activator of the MKK/JNK and p38 MAPK cascades, leading to AP-1 transcription factor activation. Related references: PMID:16893890 | PMID:9079627 | PMID:11460167 | PMID:8663074
- Independently of MAP2Ks, TAK1 promotes NF-kappa-B activation by phosphorylating IKBKB in the IKK complex upon recruitment to polyubiquitin chains via TAB2/TAB3. Related references: PMID:12589052 | PMID:8663074
- TAK1 plays a major role in osmotic stress signaling by activating MAPK8/JNK1, but not NF-kappa-B. Related references: PMID:16893890
- The protein is localized in the cytoplasm and at the cell membrane, consistent with its role in signal transduction from receptors.
- Multiple isoforms exist with tissue-specific expression: isoform 1A abundant in ovary, skeletal muscle, spleen, and blood mononuclear cells; isoform 1B in brain, kidney, small intestine; isoform 1C in prostate; isoform 1D is less abundant.
- TAK1 is subject to various post-translational modifications including phosphorylation, acetylation, and ubiquitination, which regulate its activity and signaling specificity.
- Additionally, TAK1 has roles in antiviral restriction (TRIM5 capsid-specific restriction) and in necroptosis by phosphorylating RIPK1; it phosphorylates STING1 in response to cGAMP, promoting STING1 translocation to COPII vesicles. Related references: PMID:21512573 | PMID:37832545
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 507-606) of human TAK1; this region may be less conserved, potentially contributing to species cross-reactivity with mouse. For WB, a band around 67 kDa is expected; consider using appropriate loading controls and KO-validated lysates to confirm specificity.
- In IP experiments, pre-clear lysates and optimize antibody-to-lysate ratios; the rabbit monoclonal format provides high affinity and reproducibility.
- For ELISA, the antibody can be used for capture or detection; validate with recombinant TAK1 protein or cell lysates expressing the target.
- Note that cross-reactivity is reported for human and mouse; reactivity with other species should be verified experimentally.
