Biological Background: Tafazzin / TAZ Function and Localization
- Tafazzin (TAZ), encoded by the TAFAZZIN gene and also known as protein G4.5, is a mitochondrial acyltransferase expressed as multiple isoforms via alternative splicing, with tissue-specific patterns.
- Catalyzes the remodeling of cardiolipin (CL) in the mitochondrial inner membrane through CoA-independent transacylation between phospholipids, primarily phosphatidylcholine (PC) and CL. - Required for the initiation of mitophagy, linking CL remodeling to mitochondrial quality control. Related references: PMID:33096711
- Localizes to the mitochondrial outer membrane, inner membrane, and cytoplasm, consistent with its dynamic role in phospholipid metabolism and membrane dynamics.
- Highly expressed in cardiac and skeletal muscle; mutations cause Barth syndrome (BTHS), an X-linked cardiomyopathy. Tissue-specific isoforms exist, with N-terminus-lacking variants found in leukocytes and fibroblasts but not in heart or skeletal muscle.
