Biological Background: TAF12 Function and Localization
- TAF12, also referred to as TAF2J, TAFII20, or TAFII15, is a core component of the general transcription factor TFIID, which orchestrates RNA polymerase II pre-initiation complex assembly at gene promoters. Related references: PMID:33795473
- In addition to TFIID, TAF12 is a shared subunit of the TATA-binding protein-free TAF complex (TFTC), the PCAF histone acetylase complex, and the STAGA transcription coactivator-HAT complex, linking transcription to chromatin acetylation. Related references: PMID:10373431 PMID:7729427 PMID:8598932
- TAF12 is localized to the nucleus, consistent with its role in transcription, and is expressed ubiquitously across human tissues.
- The protein is subject to multiple post-translational modifications, including phosphorylation and ubiquitin-like conjugation (e.g., isopeptide bond formation), which may modulate its activity or interactions.
- TAF12 has a calculated molecular weight of approximately 17.9 kDa, with an apparent mass of around 18 kDa on SDS-PAGE, and alternative initiation generates isoforms of varying lengths.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1-161 of human TAF12; therefore, the antibody is expected to recognize epitopes within this N-terminal fragment.
- For Western blotting, a band at approximately 18 kDa is anticipated; use appropriate molecular weight markers and validate in a human sample with verified TAF12 expression.
- This polyclonal antibody may exhibit lot-to-lot variation; it is advisable to titrate the antibody for optimal performance in your specific assay conditions.
- For ELISA, direct or sandwich formats may be optimized using the recombinant immunogen as a standard.
- When applying to other species, verify cross-reactivity experimentally, as the antibody is validated only for human TAF12.
