Biological Background: TAB3 Function and Localization
- TGF-beta-activated kinase 1 and MAP3K7-binding protein 3 (TAB3), also known as NF-kappa-B-activating protein 1 (NAP1) and TAK1-binding protein 3 (TAB-3), is an adapter protein required for the activation of JNK and NF-kappa-B signaling pathways. Related references: PMID:14633987 PMID:14766965 PMID:15327770
- It specifically recognizes Lys-63-linked polyubiquitin chains through its RanBP2-type zinc finger (NZF) domain, linking MAP3K7/TAK1 and TRAF6 to these chains. Related references: PMID:14633987 PMID:14766965 PMID:15327770
- The NZF domain recognizes polyubiquitin chains anchored to substrates like RIPK1 and RIPK2, scaffolding a signaling complex that promotes TAK1 autophosphorylation and subsequent activation of the IKK complex. Related references: PMID:15327770 PMID:18079694 PMID:22158122
- TAB3 is widely expressed and constitutively overexpressed in certain tumor tissues, suggesting a potential oncogenic role.
- The protein undergoes post-translational modifications including acetylation, methylation, and phosphorylation, and contains a coiled-coil domain, zinc-finger, and metal-binding sites.
- Alternative splicing generates multiple transcript variants, with a major and minor transcript reported.
Experimental Guidance and Technical Tips
- For Western blotting, consider using whole-cell lysates from human or mouse cells, as the antibody detects a band around 79 kDa corresponding to TAB3.
- Due to the immunogen design (N-terminal peptide, aa 1-100), ensure the epitope is accessible in your assay; denaturing conditions for WB are appropriate.
- For ELISA, validate with recombinant TAB3 protein or peptide to assess binding affinity.
- The antibody cross-reacts with mouse TAB3, so it can be used for mouse model studies; verify expression levels in the relevant tissue.
- Consider including appropriate controls (e.g., TAB3 knockout lysates) to confirm specificity.
