Biological Background: T-bet/Tbx21 Function and Localization

  • Lineage-defining transcription factor that initiates Th1 lineage development from naive Th precursor cells by activating Th1 genetic programs and repressing the opposing Th2 and Th17 genetic programs. Related references: PMID:10761931
  • Activates transcription of a set of genes important for Th1 cell function, including those encoding IFN-gamma and the chemokine receptor CXCR3, and induces permissive chromatin accessibility and CpG methylation in IFNG. Related references: PMID:33296702
  • Can activate Th1 genes via recruitment of Mediator complex and P-TEFb (composed of CDK9 and CCNT1/cyclin-T1) in the form of the super elongation complex (SEC) to super-enhancers and associated genes in activated Th1 cells. Related references: PMID:27292648
  • Inhibits the Th17 cell lineage commitment by blocking RUNX1-mediated transactivation of Th17 cell-specific transcriptinal regulator RORC, and suppresses Th2 cytokines such as IL-4, IL-5, and IL-13 via repression of GATA3 and NFATC2.
  • Protects Th1 cells from amplifying aberrant type-I IFN response in an IFN-gamma abundant microenvironment by acting as a repressor of type-I IFN transcription factors and type-I IFN-stimulated genes.
  • Localizes predominantly to the nucleus, consistent with its role as a transcription factor.
  • Exhibits T-cell specific expression; also required for the correct development of natural killer (NK) and mucosal-associated invariant T (MAIT) cells.
  • Post-translational modifications include phosphorylation, isopeptide bond formation, and ubiquitination, which may regulate its activity and stability.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 322–535 (C-terminal region) of human T-bet/Tbx21, providing specificity for the full-length protein.
  • For Western blot (WB), a band at approximately 58 kDa is expected; verify in a relevant sample such as Th1-polarized human T cells or mouse splenocytes.
  • In immunohistochemistry (IHC-P), consider heat-induced epitope retrieval (HIER) in citrate buffer (pH 6.0) for optimal detection.
  • For immunoprecipitation (IP), validate antibody performance under native conditions using nuclear extracts from activated T cells.
  • Cross-reactivity with mouse and rat T-bet allows inter-species studies; confirm detection in the appropriate tissue or cell line.
  • Titrate the antibody by ELISA to determine optimal working concentration for each specific application.
