Biological Background: Synaptotagmin-11 Function and Localization
- Synaptotagmin-11 (SYT11), also known as Synaptotagmin XI (SytXI), is a member of the synaptotagmin family involved in vesicular and membrane trafficking. Unlike many synaptotagmins, SYT11 does not bind Ca²⁺.
- SYT11 inhibits clathrin-mediated and bulk endocytosis, ensuring precision in vesicle retrieval, and plays an important role in dopamine transmission by regulating endocytosis and the vesicle-recycling process.
- SYT11 is essential for a neuronal vesicular trafficking pathway distinct from the synaptic vesicle pathway, crucial for development and synaptic plasticity.
- In macrophages and microglia, SYT11 inhibits conventional cytokine secretion (e.g., IL6 and TNF) and phagocytosis, and in astrocytes, it regulates lysosome exocytosis for membrane repair.
