- Synaptotagmin-11 (SytXI) is a member of the synaptotagmin family of vesicle-trafficking proteins; unlike many family members, it does not bind Ca2+ and is calcium-independent in its functions.
- It acts as an inhibitor of clathrin-mediated and bulk endocytosis, helping to ensure precision in vesicle retrieval, and is critical for dopamine transmission by regulating endocytosis and synaptic vesicle recycling.
- SytXI is an essential component of a neuronal vesicular trafficking pathway distinct from the canonical synaptic vesicle pathway, playing a key role in neuronal development and synaptic plasticity.
- In non-neuronal cells, it inhibits conventional cytokine secretion (e.g., IL6 and TNF) and phagocytosis in macrophages and microglia, while in astrocytes it regulates lysosome exocytosis for plasma membrane repair.
