Biological Background: Synaptophysin Function and Localization
- Synaptophysin, also known as major synaptic vesicle protein p38, is encoded by the SYP gene. Multiple isoforms are produced by alternative splicing.
- It plays a key role in organizing membrane components and targeting synaptic vesicles to the plasma membrane, thereby regulating both short-term and long-term synaptic plasticity (By similarity).
- This integral membrane protein is a member of the synaptophysin family and contains four transmembrane helices, contributing to vesicle structure and trafficking.
- Subcellularly, it is localized to the cytoplasmic side of synaptic vesicle membranes and is enriched at synapses and synaptosomes.
- Post-translational modifications include glycosylation, phosphorylation, and ubiquitination. It contains calcium-binding sites and disease-associated variants linked to intellectual disability.
