Biological Background: SYCP3 Function and Localization
- Synaptonemal complex protein 3 (SYCP3), also known as SCP3, COR1, SPGF4, or RPRGL4, is a 27729 Da testis-specific protein encoded by the SYCP3 gene (Gene ID 50511) in humans.
- Component of the synaptonemal complex (SC), a tripartite protein structure formed between homologous chromosomes during meiotic prophase, essential for proper chromosome synapsis and recombination.
- Required for centromere pairing during meiosis in male germ cells (By similarity), facilitating correct chromosome segregation.
- Indispensable for normal meiosis during spermatogenesis and male fertility (PubMed:14643120), while playing a lesser role in female fertility. Related references: PMID:14643120
- Acts upstream of HORMAD1 and HORMAD2 phosphorylation (By similarity), a critical step in meiotic checkpoint signaling.
- Localizes to the nucleus, specifically associating with chromosomes and centromeres during meiotic prophase.
- Contains coiled-coil domains and exhibits DNA-binding activity, with its 3D structure determined, classifying it as a phosphoprotein.
Experimental Guidance and Technical Tips
- Immunogen design: The immunogen corresponds to the N-terminal region (1-236) of human SYCP3, which shows high conservation across human, mouse, and rat, supporting cross-reactivity.
- Western blot: Due to testis-specific expression, consider using human, mouse, or rat testis tissue lysates. The predicted molecular weight is approximately 28 kDa.
- ELISA: The antibody may be suitable for direct or indirect ELISA; optimization of coating antigen and antibody dilution is recommended.
- Cross-reactivity: Validated reactivity with mouse and rat, but verify in the relevant sample system.
