Biological Background: SV2A Function and Localization
- SV2A (Synaptic vesicle glycoprotein 2A) is a synaptic vesicle membrane protein that belongs to the SV2 family of transporters. It features twelve transmembrane domains and acts as a neurotransmitter transporter.
- It plays a critical role in the control of regulated secretion in neural and endocrine cells, enhancing low-frequency neurotransmission by positively regulating vesicle fusion and maintaining the readily releasable pool of synaptic vesicles (By similarity).
- SV2A localizes to the presynapse and the membranes of synaptic vesicles and secretory vesicles, consistent with its role in neurotransmitter release.
- The protein is glycosylated, which enhances its function as a receptor for botulinum neurotoxin type A2 (BoNT/A); unglycosylated SV2A still interacts but with lower affinity. Related references: PMID:29649119
- SV2A is associated with epilepsy based on disease variant annotations, and alternative splicing produces multiple isoforms.
- Post-translational modifications include phosphorylation, and the protein serves as a key synaptic marker in studies of neurotransmission and neurological disorders.
- The theoretical molecular weight of the unmodified human SV2A is approximately 82.7 kDa.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 356–447, which corresponds to a cytoplasmic loop region of SV2A. This design may enable the antibody to detect both native and denatured protein in Western blotting under reducing conditions.
- For WB analysis, consider using lysates from brain or neural tissues (human, mouse, rat) where SV2A expression is high. Validate the antibody in your specific sample system to confirm specificity and sensitivity.
- In ELISA applications, the immunogen can serve as a positive control coat antigen. Perform titration experiments to optimize the antibody concentration for your assay setup.
- Cross-reactivity with mouse and rat has been observed, allowing detection across these species. However, sequence homology suggests potential reactivity with other species; validation is recommended.
- As a polyclonal antibody, lot-to-lot consistency should be assessed, and blocking with serum or BSA should be optimized to reduce background.
