Biological Background: Survivin Function and Localization

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the full-length survivin (aa 1–142) sequence, making this antibody suitable for detecting both wild-type survivin and its isoforms. Because the predicted molecular weight is ~16 kDa but the observed band may range 8–18 kDa, consider including positive and negative controls and validating the band pattern in your specific sample system.
  • For western blotting, use a reducing agent and boil samples appropriately. A band around 16–18 kDa is expected in lysates from proliferating cancer cells (e.g., HeLa, Jurkat) or fetal tissues, but expression may be low in normal adult tissues.
  • For ELISA applications, pairing with a suitable capture or detection antibody that recognizes a different epitope is recommended. Validate the antibody pair using recombinant survivin protein or cell lysates with known expression levels.
  • Cross-reactivity with mouse and rat orthologs allows flexible model system use; however, confirm reactivity in your experimental context with appropriate species-specific controls.
