Biological Background: SUPV3L1 Function and Localization

  • ATP-dependent RNA helicase SUPV3L1, also known as suppressor of var1 3-like protein 1 (SUV3-like protein 1). It belongs to the helicase family and contains a transit peptide for mitochondrial import.
  • Major helicase player in mitochondrial RNA metabolism. Component of the mitochondrial degradosome (mtEXO) complex, that degrades 3' overhang double-stranded RNA with a 3'-to-5' directionality in an ATP-dependent manner. Involved in the degradation of non-coding mitochondrial transcripts (MT-ncRNA) and tRNA-like molecules (PubMed:29967381). Related references: PMID:29967381
  • Exhibits ATP-dependent multisubstrate helicase activity, able to unwind double-stranded DNA and RNA, and RNA/DNA heteroduplexes in the 5'-to-3' direction.
  • Plays a role in the mitochondrial RNA surveillance system, regulating the stability of mature mRNAs, removing aberrantly formed mRNAs, and rapidly degrading non-coding processing intermediates.
  • Implicated in recombination and chromatin maintenance pathways. May protect cells from apoptosis and associates with mitochondrial DNA.
  • Subcellular localization: found in the nucleus, mitochondrial matrix, and mitochondrial nucleoid.
  • Broadly expressed across tissues, as indicated by tissue specificity data.
  • Post-translational modifications include acetylation and phosphorylation. Keywords: 3D-structure, Acetylation, ATP-binding, Helicase, Hydrolase, Mitochondrion, Mitochondrion nucleoid, Nucleotide-binding, Nucleus, Phosphoprotein, Proteomics identification, Reference proteome, Transit peptide.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (aa 577-786) of human SUPV3L1, which may enhance specificity for the target protein. Validate in the relevant sample system to confirm expected reactivity.
  • For Western blot (WB), a band around 88 kDa is expected based on the predicted molecular weight. Consider using appropriate loading controls and mitochondrial fractionation if needed.
  • In immunofluorescence/immunocytochemistry (IF/ICC), the protein localizes to mitochondria and nucleus; co-staining with organelle markers is recommended to confirm localization.
  • The antibody cross-reacts with mouse and rat, enabling studies across these species. However, always verify cross-reactivity in your experimental setup.
  • For ELISA applications, consider optimizing antibody concentration and assay conditions for your specific antigen detection needs.
  • Given the protein's involvement in mitochondrial RNA metabolism, experimental conditions such as mitochondrial isolation or specific inhibitors may be explored to modulate its activity.
