- Transcription elongation factor SPT6 (SPT6H) acts as a histone H3-H4 chaperone, facilitating RNA polymerase II transcription elongation and enhancing Tat-mediated activation of HIV-1 transcription.
- Forms a complex with IWS1 and the C-terminal domain (CTD) of RNAPII subunit RPB1, coupling transcription with mRNA processing, splicing, and export.
- Recruits mRNA export factors (ALYREF/THOC4, EXOSC10) and histone modifying enzymes (e.g., SETD2) to ensure proper mRNA maturation and deposition of elongation-coupled H3K36me3 marks.
- Associates with SETD1A to catalyze H3K4me3 at AICDA target loci, regulating immunoglobulin class-switch recombination and somatic hypermutation.
- Promotes muscle differentiation by stabilizing the chromatin interaction of H3K27 demethylase KDM6A, thereby erasing repressive H3K27me3 marks at myogenic genes.
- Localizes to the nucleus and is ubiquitously expressed across tissues.
