Biological Background: SUPT5H/SPT5 Function and Localization
- SUPT5H/SPT5 encodes the large subunit (p160) of the DRB sensitivity-inducing factor (DSIF) complex, a transcription elongation factor that regulates RNA polymerase II processivity and mRNA processing. Related references: PMID:10075709 PMID:10199401 PMID:10421630

Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 125–425 within the mid-region of human SUPT5H. This region is conserved in mouse, supporting the observed cross-reactivity.
- For western blot (WB) analysis, a predominant band at approximately 121 kDa is expected. Consider using nuclear extracts to enrich endogenous target detection.
- Because SUPT5H forms a stable complex with SUPT4H (DSIF), co-immunoprecipitation (IP) experiments can be designed to study the DSIF complex and its associated factors.
- The antibody is also suitable for ELISA; however, optimal working conditions should be validated in the specific assay system.
