  • SUPT16H (SPT16) is a subunit of the FACT (Facilitates Chromatin Transcription) complex, a general chromatin factor that reorganizes nucleosomes to facilitate DNA-templated processes such as transcription elongation, DNA replication, and DNA repair.
  • During transcription elongation, FACT acts as a histone chaperone, destabilizing nucleosomes by promoting the dissociation of one histone H2A-H2B dimer, thereby enabling RNA polymerase II passage, and then restoring nucleosome structure afterward.
  • The FACT complex is also implicated in the phosphorylation of p53/TP53 at Ser-392 via its association with casein kinase II (CK2).
  • SUPT16H is localized to the nucleus and associates with chromosomes during active DNA transactions.
  • The protein is ubiquitously expressed across tissues.
  • Post-translational modifications include acetylation, ADP-ribosylation, phosphorylation, and ubiquitylation (isopeptide bond formation).
