Biological Background: SUPT16H (SPT16) Function and Localization
- SUPT16H (SPT16) is a subunit of the FACT (Facilitates Chromatin Transcription) complex, a general chromatin factor that reorganizes nucleosomes to facilitate DNA-templated processes such as transcription elongation, DNA replication, and DNA repair.
- During transcription elongation, FACT acts as a histone chaperone, destabilizing nucleosomes by promoting the dissociation of one histone H2A-H2B dimer, thereby enabling RNA polymerase II passage, and then restoring nucleosome structure afterward.
- The FACT complex is also implicated in the phosphorylation of p53/TP53 at Ser-392 via its association with casein kinase II (CK2).
- SUPT16H is localized to the nucleus and associates with chromosomes during active DNA transactions.
- The protein is ubiquitously expressed across tissues.
- Post-translational modifications include acetylation, ADP-ribosylation, phosphorylation, and ubiquitylation (isopeptide bond formation).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 520-695) of human SUPT16H; consider verifying antibody specificity against full-length protein in your sample system.
- For Western blot, the predicted molecular weight is ~120 kDa; use a protein ladder with appropriate range and optimize transfer conditions for high molecular weight proteins.
- For immunofluorescence/immunocytochemistry, validate fixation and permeabilization protocols to ensure nuclear antigen accessibility.
- Cross-reactivity with mouse and rat has been demonstrated; if using other species, check sequence homology before testing.
