Biological Background: SUN2 Function and Localization

  • SUN2, also known as protein unc-84 homolog B, Rab5IP, and Sad1/unc-84 protein-like 2, is encoded by the SUN2 gene and exists in multiple isoforms through alternative splicing.
  • As a core component of the LINC (LInker of Nucleoskeleton and Cytoskeleton) complex, SUN2 bridges the nuclear lamina and the cytoskeleton, transmitting mechanical forces across the nuclear envelope and governing nuclear positioning and movement.
  • SUN2 collaborates with SYNE2 to form transmembrane actin-associated nuclear (TAN) lines that couple the nucleus to retrograde F-actin flow, essential for actin-dependent nuclear migration.
  • During development, SUN2 is required for interkinetic nuclear migration (INM) and radial neuronal and glial migration in the cerebral cortex, as well as nuclear migration in retinal photoreceptor progenitors via dynein-dynactin and kinesin motors.
