Biological Background: SUMO4 Function and Localization

  • SUMO4 is a ubiquitin-like protein that is covalently attached to target lysines as a monomer, modulating protein localization, stability, or activity.
  • Under oxidative stress, SUMO4 conjugates to various antioxidant enzymes, chaperones, and stress defense proteins.
  • SUMO4 negatively regulates transcriptional activity of NFKBIA, TFAP2A, and FOS, and positively regulates NR3C1.
  • Covalent attachment requires prior activation by the E1 complex SAE1-SAE2 and linkage to the E2 enzyme UBE2I.
  • Expressed mainly in adult and embryonic kidney, with highest immune tissue expression in lymph node and spleen.
  • Associated with diabetes mellitus (IDDM5); contains isopeptide bonds and participates in the Ubl conjugation pathway.

Experimental Guidance and Technical Tips

  • For Western blot (WB), the predicted molecular weight is 11 kDa. Use reducing conditions and consider positive controls such as HEK293 cell lysates or immune tissue extracts.
  • For immunohistochemistry (IHC-P), validate in kidney or lymph node sections, as these tissues show high SUMO4 expression.
