Biological Background: SUMO4 Function and Localization
- SUMO4 is a ubiquitin-like protein that is covalently attached to target lysines as a monomer, modulating protein localization, stability, or activity.
- Under oxidative stress, SUMO4 conjugates to various antioxidant enzymes, chaperones, and stress defense proteins.
- SUMO4 negatively regulates transcriptional activity of NFKBIA, TFAP2A, and FOS, and positively regulates NR3C1.
- Covalent attachment requires prior activation by the E1 complex SAE1-SAE2 and linkage to the E2 enzyme UBE2I.
- Expressed mainly in adult and embryonic kidney, with highest immune tissue expression in lymph node and spleen.
- Associated with diabetes mellitus (IDDM5); contains isopeptide bonds and participates in the Ubl conjugation pathway.
Experimental Guidance and Technical Tips
- For Western blot (WB), the predicted molecular weight is 11 kDa. Use reducing conditions and consider positive controls such as HEK293 cell lysates or immune tissue extracts.
- For immunohistochemistry (IHC-P), validate in kidney or lymph node sections, as these tissues show high SUMO4 expression.
- For immunofluorescence (IF/ICC), confirm subcellular localization, considering that SUMO4 conjugation may alter target distribution.
- Due to the small size, use appropriate gel percentage and transfer conditions; low molecular weight markers are recommended.
- Cross-reactivity with human, mouse, and rat has been reported; validate in the relevant species model.
