Biological Background: SUMO4 Function and Localization
- SUMO4 (Small ubiquitin-related modifier 4) is a ubiquitin-like protein that can be covalently attached to target lysines as a monomer.
- Unlike ubiquitin, SUMO4 conjugation does not lead to protein degradation; instead, it modulates substrate subcellular localization, stability, or activity.
- Upon oxidative stress, SUMO4 conjugates to various anti-oxidant enzymes, chaperones, and stress defense proteins, suggesting a role in cellular stress response.
- SUMO4 negatively regulates the transcriptional activity of NFKBIA, TFAP2A, and FOS, while positively regulating NR3C1 transcriptional activity.
- Covalent attachment to substrates requires activation by the E1 complex SAE1-SAE2 and linkage to the E2 enzyme UBE2I, forming an isopeptide bond.
- Tissue expression is mainly in adult and embryonic kidney, with highest levels in immune tissues such as lymph node and spleen.
- SUMO4 is associated with diabetes mellitus (IDDM5) and is part of the ubiquitin-like conjugation pathway.

- The immunogen is a peptide from residues 40-95, which may recognize both monomeric and conjugated SUMO4; consider assessing specificity for sumoylated versus free forms in your assay.
- For Western blot, SUMO4 has a predicted molecular weight of ~11 kDa, but covalent conjugates will migrate at higher molecular weights; use appropriate reducing conditions and validate with known sumoylation substrates.
- For immunohistochemistry (IHC-P), positive staining is expected in kidney and immune tissues; include appropriate negative controls to confirm specificity.
- For ELISA, optimize coating and detection conditions using purified SUMO4 or recombinant proteins as standards.
