Biological Background: SUMO3 Function and Localization
- SUMO3 (Small ubiquitin-related modifier 3) is a ubiquitin-like protein, also known as SUMO-2, SMT3A, or Smt3B, encoded by the SUMO3 gene.
- It can be covalently attached to target lysines either as a monomer or as a lysine-linked polymer, and does not mainly target proteins for degradation, potentially functioning as an antagonist of ubiquitin.
- The conjugation of SUMO3 to substrates requires the sequential action of the E1 complex SAE1-SAE2, the E2 enzyme UBE2I, and can be enhanced by E3 ligases such as PIAS1-4, RANBP2, or CBX4. Related references: PMID:11451954, PMID:18538659, PMID:21965678
- SUMO3 plays a role in the regulation of sumoylation status of SETX, linking it to DNA repair and neurological function pathways. Related references: PMID:24105744
- It is involved in various cellular processes including nuclear transport, DNA replication and repair, mitosis, and signal transduction.
- SUMO3 localizes to the cytoplasm, nucleus, and PML bodies, with predominant expression in liver tissue.
- The mature protein has a molecular weight of approximately 11.6 kDa, consistent with its small modifier function, and belongs to the SUMO family of ubiquitin-like modifiers.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 1-100 of human SUMO3 (NP_008867.2). For validation, consider using recombinant full-length SUMO3 or cell lysates expressing the protein.
- Recommended applications include Western blot (WB), immunohistochemistry on paraffin sections (IHC-P), and ELISA. Dilutions and conditions should be optimized experimentally for your specific samples.
- The antibody shows cross-reactivity with human, mouse, and rat, making it a candidate for studies in rodent models; verify reactivity in your model system.
- In WB, expect bands at ~12 kDa (free SUMO3) as well as higher molecular weight bands corresponding to sumoylated conjugates. Include appropriate positive and negative controls.
