Biological Background: SUMO3 Function and Localization
- SUMO3 (Small Ubiquitin-related Modifier 3) is a ubiquitin-like protein encoded by the SUMO3 gene (GeneID: 6612). It is a member of the SUMO family and is also known as SMT3A, Smt3B, SMT3H1, or SUMO-2.
- Unlike ubiquitin, SUMO3 primarily functions as a post-translational modifier that does not target proteins for degradation; instead, it may antagonize ubiquitin-dependent proteolysis. It is covalently attached to target lysines as a monomer or polymer.
- The conjugation process requires an E1 activating enzyme (SAE1-SAE2), an E2 conjugating enzyme (UBE2I), and is promoted by E3 ligases such as PIAS1-4, RANBP2, or CBX4. Related references: PMID:11451954, PMID:18538659, PMID:21965678
- SUMO3 participates in nuclear transport, DNA replication and repair, mitosis, and signal transduction. It is also critical for regulating the sumoylation of SETX. Related references: PMID:24105744
- Subcellularly, SUMO3 is found in the cytoplasm, nucleus, and PML nuclear bodies.

Experimental Guidance and Technical Tips
- Due to the small size of SUMO3 (~12 kDa), use 15% SDS-PAGE gels or gradient gels for optimal resolution in Western blot, and ensure efficient transfer onto PVDF membranes with low molecular weight cut-offs.
- For immunofluorescence (IF/ICC), fix cells with 4% paraformaldehyde and permeabilize with 0.1-0.2% Triton X-100 to allow antibody access to nuclear and PML body-localized SUMO3.
- Validate antibody specificity in relevant cell lines or tissues; SUMO3 is highly expressed in liver, so HepG2 or primary hepatocytes may serve as positive controls.
- Since this polyclonal antibody may recognize both free and conjugated forms, consider using 1% SDS or other denaturing conditions to reduce non-specific binding in ELISA.
