Biological Background: SUMO2/3 Function and Localization
- Small ubiquitin-related modifier 2 (SUMO2), also known as HSMT3, SMT3B, Sentrin-2, or Ubiquitin-like protein SMT3B, is a member of the SUMO family of ubiquitin-like proteins. It shares high sequence homology with SUMO3, and this antibody recognizes both SUMO2 and SUMO3 (SUMO2/3).
- SUMO2 functions as a ubiquitin-like protein that can be covalently attached to lysine residues of target substrates, either as a monomer or as a lysine-linked polymeric chain. This modification, termed SUMOylation, requires activation by the E1 complex SAE1-SAE2, conjugation by the E2 enzyme UBE2I, and often promotion by E3 ligases such as PIAS1-4, RANBP2, CBX4, or ZNF451. - The immunogen used corresponds to amino acids 1-95 of human SUMO2/3, covering the full-length protein of ~11 kDa. When performing Western blot, expect a band around 11 kDa for free SUMO2/3, with additional higher molecular weight bands representing SUMOylated protein conjugates.
- For immunohistochemistry (IHC-P), consider standard antigen retrieval methods and validate the antibody in relevant tissue samples, as SUMO2 is broadly expressed but may show nuclear enrichment.
- In ELISA applications, use appropriate controls to confirm specificity for SUMO2/3, given the high homology between SUMO family members.
