Biological Background: SUMO2/3 Function and Localization

  • Small ubiquitin-related modifier 2 (SUMO2), also known as HSMT3, SMT3B, SUMO-3, Sentrin-2, is a ubiquitin-like protein that can be covalently attached to lysine residues of target proteins as a monomer or as a lysine-linked polymer.
  • Covalent attachment of SUMO2 to substrates requires prior activation by the E1 complex SAE1-SAE2, linkage to the E2 enzyme UBE2I, and can be promoted by E3 ligases such as PIAS1-4, RANBP2, CBX4 or ZNF451. Related references: PMID:26524494
  • SUMO2 modification plays a crucial role in a number of cellular processes including nuclear transport, DNA replication and repair, mitosis, and signal transduction.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal region (aa 1-95) of human SUMO2/3. Note that SUMO proteins are often conjugated to target proteins, so the antibody may detect free SUMO (~11 kDa) as well as higher molecular weight SUMOylated proteins in Western blot. Validate the band pattern in your specific sample system.
  • For immunofluorescence, expect nuclear staining with possible punctate PML body localization. Optimize fixation and permeabilization conditions for your cell type.
  • The antibody cross-reacts with human, mouse, and rat. Confirm reactivity and specificity in your target tissue or cell line before large-scale experiments.
  • For ELISA, consider coating with recombinant SUMO2/3 and titrate the antibody to determine an optimal working dilution for your assay format.
