Biological Background: SUMO1 Function and Localization

  • Small ubiquitin-related modifier 1 (SUMO1) is a ubiquitin-like protein that becomes covalently conjugated to target proteins, a modification known as SUMOylation. It is also designated GMP1, PIC1, SMT3C, UBL1, and Sentrin.
  • SUMOylation requires prior activation by the E1 complex SAE1-SAE2, linkage to the E2 enzyme UBE2I, and often promotion by E3 ligases such as PIAS1-4, RANBP2, or CBX4. This reversible modification regulates nuclear transport, DNA replication and repair, mitosis, and signal transduction.
  • A key substrate is RANGAP1, whose SUMO1 conjugation targets it to the nuclear pore complex protein RANBP2. SUMO1 also modulates the gating characteristics of the voltage-gated potassium channel KCNB1. Related references: PMID:19223394

  • Polymeric SUMO1 chains can be polyubiquitinated, marking modified proteins for proteasomal degradation. SUMO1 may also regulate genes involved in palate development.
  • Subcellular localization includes the nucleus (membrane, speckles, PML bodies), cytoplasm, and cell membrane, reflecting its diverse substrate repertoire.
  • Post-translational modifications of SUMO1 itself include acetylation, phosphorylation, and isopeptide bond formation. Alternative splicing generates additional isoforms.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal region (aa 1–101) of human SUMO1 (P63165); this region is likely to recognize all splicing isoforms. Validate isoform specificity in your system if needed.
  • For Western blot, unconjugated SUMO1 migrates at ~12 kDa. SUMOylated substrates appear at higher masses; use appropriate controls (e.g., SUMO deconjugating enzymes or knockout lysates) to confirm specificity.
  • In IHC-P and IF/ICC, signal may be nuclear, cytoplasmic, or membrane-associated depending on the target protein’s SUMOylation status. Validate with positive and negative tissue/cell controls.
  • For ELISA applications, the antibody can be used as a capture or detection reagent after empirical titer and pairing optimization.
  • Store antibody at -20°C or as recommended upon receipt; avoid repeated freeze-thaw cycles.
