Biological Background: SUMF2 Function and Localization
- SUMF2, also known as pFGE (paralog of formylglycine-generating enzyme) or sulfatase-modifying factor 2, is an inactive paralog of SUMF1, the enzyme responsible for converting inactive sulfatases to their active form.
- It lacks formylglycine generating activity and cannot activate newly synthesized sulfatases; instead, it inhibits sulfatase activation by SUMF1.
- The protein localizes to the lumen of the endoplasmic reticulum, where post-translational modification of sulfatases occurs.
- SUMF2 is widely expressed in human tissues, with highest levels detected in kidney, liver, and placenta.
- Key post-translational modifications include glycosylation and disulfide bond formation; a calcium-binding site is also predicted.
- A three-dimensional structure is available (UniProt keyword: 3D-structure), providing insights into its interaction with SUMF1 and sulfatases.
Experimental Guidance and Technical Tips
- The immunogen used (aa 45-320) corresponds to the mature SUMF2 protein, likely recognizing native conformations in WB and IF/ICC.
- For Western blot, a band around 34 kDa is expected; validate in lysates from tissues with high expression (e.g., kidney, liver, placenta).
- In IF/ICC, permeabilization is required to access the ER lumen; consider co-staining with an ER marker to confirm localization.
- Cross-reactivity with mouse and rat has been reported; however, sequence alignment of the immunogen region is advisable for other species.
- The antibody is also suitable for ELISA; optimal coating and detection conditions should be determined empirically.
