Biological Background: SULT2B1 Function and Localization
- SULT2B1, a member of the cytosolic sulfotransferase family, is encoded by the SULT2B1 gene and exists in tissue-specific isoforms via alternative splicing.
- It catalyzes the transfer of a sulfonate group from 3'-phospho-5'-adenylyl sulfate (PAPS) to hydroxyl groups of substrates, primarily sulfating cholesterol and hydroxysteroids like pregnenolone and dehydroepiandrosterone (DHEA). Related references: PMID:12145317, PMID:16855051, PMID:21855633
- Plays a critical role in epidermal cholesterol metabolism and regulation of keratinocyte proliferation/differentiation, as evidenced by expression at the stratum granulosum-stratum corneum junction. Related references: PMID:28575648
- Localized to the cytoplasm, microsome, and nucleus, enabling sulfation activity in multiple cellular compartments.
- Expressed at high levels in placenta, prostate, and trachea, with lower levels in small intestine and lung. Related references: PMID:9799594
- Post-translationally modified by phosphorylation, and genetic variations are linked to the skin disorder ichthyosis.
Experimental Guidance and Technical Tips
- Based on the full-length immunogen, this rabbit polyclonal antibody is expected to recognize multiple epitopes, potentially enabling detection of both native and denatured SULT2B1 protein in applications such as WB and IF/ICC.
- For WB, a band approximately 41 kDa is observed; loading controls should be included, and validation in human, mouse, or rat tissue lysates (e.g., placenta or prostate) is recommended.
- For IF/ICC, fixation and permeabilization conditions should be optimized, and nuclear or cytoplasmic staining may be observed consistent with SULT2B1 localization.
- ELISA applications may require titration of the capture/detection antibody concentrations; consider pairing with a validated SULT2B1 standard protein.
