Biological Background: SULT2A1 Function and Localization
- SULT2A1 is a sulfotransferase that uses 3'-phospho-5'-adenylyl sulfate (PAPS) as a sulfonate donor to catalyze the sulfonation of a wide range of steroids and sterols, including pregnenolone, androsterone, dehydroepiandrosterone (DHEA), bile acids, cholesterol, and many xenobiotics with alcohol or phenol groups (PubMed:14573603, PubMed:18042734, PubMed:19589875). Related references: PMID:14573603 PMID:18042734 PMID:19589875
- Plays an important role in maintaining steroid and lipid homeostasis, as sulfonation increases water solubility and renal excretion of steroids (PubMed:14573603, PubMed:19589875, PubMed:21187059). Related references: PMID:14573603 PMID:19589875 PMID:21187059
- Key enzyme in bile acid metabolism, mediating the formation of 3-sulfated bile acids, which are major components of sulfate-conjugated bile acids in human urine (PubMed:2268288, PubMed:20102295). Related references: PMID:2268288 PMID:20102295
- Also catalyzes the metabolic activation of potent carcinogenic polycyclic arylmethanols via sulfonation (By similarity).
- Subcellular localization: cytoplasm.
- Tissue specificity: highly expressed in liver and adrenal gland, with lower levels in kidney; expression is higher in fetal adrenal than in fetal liver or kidney.
- Belongs to the sulfotransferase family; also known as hydroxysteroid sulfotransferase (HST), DHEA-ST, and bile salt sulfotransferase, with multiple aliases including ST2, STD, hSTa, ST2A1, ST2A3, and DHEA-ST8.
Experimental Guidance and Technical Tips
- For Western blotting, the predicted molecular weight is approximately 34 kDa; consider using a positive control such as human liver or adrenal tissue lysate.
- For immunohistochemistry-paraffin sections (IHC-P), optimize antigen retrieval and validate staining in relevant tissues including liver and adrenal gland, where SULT2A1 expression is high.
- This antibody has been shown to cross-react with mouse and rat; validate in your sample system if using non-human models.
- ELISA applications are supported; ensure appropriate antigen coating and detection conditions are optimized for your assay format.
