Biological Background: SULT2A1 Function and Localization
- SULT2A1 is a sulfotransferase that catalyzes the transfer of a sulfonate group from 3'-phospho-5'-adenylyl sulfate (PAPS) to various steroids and bile acids, playing a critical role in hormone and lipid homeostasis. Related references: PMID:14573603, PMID:18042734, PMID:19589875, PMID:21187059
- It mediates the sulfation of a wide range of endogenous and xenobiotic compounds, including DHEA, pregnenolone, androsterone, cholesterol, and various alcohols and phenols. Related references: PMID:14573603, PMID:18042734, PMID:19589875
- SULT2A1 is essential for bile acid metabolism, catalyzing the formation of 3-sulfated bile acids that have increased water solubility for renal excretion. Related references: PMID:2268288, PMID:20102295
- This enzyme is predominantly expressed in the liver and adrenal glands, with lower levels in the kidney; during fetal development, adrenal expression surpasses hepatic and renal expression.
- Subcellularly, SULT2A1 localizes to the cytoplasm.
- Post-translational modifications include phosphorylation, as identified by proteomics.
- SULT2A1 is also known by multiple aliases, including HST, ST2, STD, hSTa, DHEAS, ST2A1, ST2A3, DHEA-ST, and SULT2A3.
Experimental Guidance and Technical Tips
- For Western blot, human liver or adrenal tissue lysates are recommended as positive controls; a specific band at approximately 34 kDa corresponding to SULT2A1 may be detected. Optimize antibody dilution and exposure times empirically.
- For ELISA, consider coating with recombinant SULT2A1 protein or using cell lysates, and validate detection with a standard curve. Cross-species reactivity with mouse suggests potential use in mouse model studies.
- The immunogen comprising amino acids 1-145 of human SULT2A1 may confer epitope specificity toward the N-terminal region; verify cross-reactivity with other sulfotransferase family members if needed.
