Biological Background: Sulfotransferase 1E1 Function and Localization

  • SULT1E1 (Sulfotransferase 1E1), also known as estrogen sulfotransferase (EST-1), is a key enzyme in estrogen homeostasis. It catalyzes the sulfate conjugation of estradiol and estrone using 3'-phospho-5'-adenylyl sulfate (PAPS) as a sulfonate donor, leading to their inactivation. Related references: PMID:11006110 PMID:11884392 PMID:7779757

  • SULT1E1 also sulfates other substrates including dehydroepiandrosterone (DHEA), pregnenolone, (24S)-hydroxycholesterol, and xenobiotic compounds like ethinylestradiol and diethylstilbestrol, albeit at lower efficiency. It does not act on cortisol, testosterone, or dopamine. Related references: PMID:11006110 PMID:19589875

  • The enzyme may contribute to gut microbiota-host metabolic interactions by sulfonating 4-ethylphenol (4-EP), a gut bacteria-derived metabolite. The product 4-EPS can cross the blood-brain barrier and potentially affect brain function and myelination.

  • SULT1E1 is predominantly expressed in the liver, intestine, and at lower levels in the kidney, consistent with its role in hormone and xenobiotic metabolism.

  • The protein localizes to the cytoplasm, particularly the cytosol, where it exerts its sulfotransferase activity.

  • SULT1E1 belongs to the sulfotransferase 1 family and possesses both lipid-binding and steroid-binding domains, reflecting its substrate specificity.

  • Post-translational modifications and structural features have been characterized (3D-structure available), and it is identified in proteomics studies as a reference proteome component.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant fragment corresponding to amino acids 140-294 of human SULT1E1; this region may contribute to specific recognition of the target in Western blot, IF/ICC, and ELISA applications.
  • For Western blot, a band at approximately 35 kDa is anticipated; however, endogenous expression levels and post-translational modifications may affect the observed molecular weight. Validate in the relevant sample system.
  • In immunofluorescence and immunocytochemistry, staining consistent with cytoplasmic localization is expected. Appropriate fixation and permeabilization methods should be optimized.
  • For ELISA, the antibody can be considered as a capture or detection reagent, but optimal dilutions and pairings should be experimentally determined.
