Biological Background: SULT1C4 Function and Localization
- Sulfotransferase 1C4 (SULT1C4), also known as SULT1C2, is a member of the sulfotransferase 1 family that catalyzes the transfer of a sulfonate group from 3'-phospho-5'-adenylyl sulfate (PAPS) to phenolic compounds.
- It plays a key role in the metabolism of xenobiotics, including dietary flavonoids (phytoestrogens) and environmental estrogens like bisphenol A. These substrates are sulfated more efficiently than 17beta-estradiol. Related references: PMID:17425406 PMID:26948952 PMID:28222028
- SULT1C4 can also sulfate anthracycline drugs such as doxorubicin and its analog epirubicin, suggesting a role in modulating the efficacy and toxicity of these antitumor agents. Related references: PMID:26948952
- The enzyme is predominantly localized in the cytoplasm/cytosol, consistent with its function in Phase II detoxification.
- It is highly expressed in fetal lung and kidney, with lower expression detected in fetal heart, adult kidney, ovary, and spinal cord.
- Alternative splicing of the SULT1C4 gene generates multiple transcript variants, potentially expanding functional diversity.
- The predicted molecular weight of the canonical isoform is approximately 35.5 kDa, while this antibody detects a band at ~36 kDa in denaturing Western blots.
Experimental Guidance and Technical Tips
- For Western blotting, start with a standard dilution range and optimize based on sample type and expression level; a band near 36 kDa is expected.
- As a polyclonal antibody, lot-to-lot consistency should be verified for critical quantitative assays; consider performing a bridging study when switching lots.
- Cross-reactivity with mouse and rat SULT1C4 homologs makes this antibody suitable for studies in common rodent models.
- For immunofluorescence/immunocytochemistry, fixation and permeabilization conditions should be empirically determined; mild fixation (e.g., 4% PFA) is often effective for cytosolic proteins.
- ELISA protocols can be developed using either direct antigen coating or a capture format; validate the assay with a known positive control.
