Biological Background: SULT1A3 Function and Localization
- Sulfotransferase that utilizes 3'-phospho-5'-adenylyl sulfate (PAPS) as a sulfonate donor to catalyze the sulfate conjugation of phenolic monoamines (dopamine, (R)-adrenaline/epinephrine, (R)-noradrenaline/norepinephrine and serotonin) and phenolic/catechol drugs. - Catalyzes the sulfation of thyroid hormones T4, T3, rT3, and 3,3'-T2, with substrate preference 3,3'-T2 > rT3 > T3 > T4. Related references: PMID:10199779
- Localized in the cytoplasm; expressed in liver, colon, kidney, lung, brain, spleen, small intestine, placenta, and leukocytes.
- Also known as ST1A3, STM, HAST3, M-PST, and TL-PST; involved in catecholamine, lipid, and steroid metabolism.
- Alternative splicing generates multiple isoforms; belongs to the sulfotransferase 1 family.
- Direct protein sequencing and 3D-structure determination have been reported.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1–100), which is conserved across human, mouse, and rat, consistent with reported cross-reactivity; perform species-specific validation if required.
- In Western blot, expect a primary band at approximately 34 kDa; liver or colon lysates may yield robust signals.
- For immunofluorescence/immunocytochemistry, SULT1A3 exhibits cytoplasmic staining; permeabilization is necessary.
- ELISA applications may benefit from checkerboard titrations to optimize antibody and antigen concentrations.
- Not recommended for applications beyond WB, IF/ICC, and ELISA without further empirical validation.
