Biological Background: SULF1 Function and Localization
- SULF1 (Extracellular sulfatase Sulf-1) is also known as arylsulfatase or N-acetylglucosamine-6-sulfatase, encoded by the SULF1 gene. It is a member of the sulfatase family and functions as a heparin-degrading endosulfatase.
- Exhibits arylsulfatase activity and highly specific endoglucosamine-6-sulfatase activity, removing sulfate from the C-6 position of glucosamine within specific subregions of intact heparin (PubMed:12368295, PubMed:12686563).
Related references: PMID:12368295 PMID:12686563 - Diminishes heparan sulfate proteoglycan (HSPG) sulfation, thereby inhibiting signaling by heparin-dependent growth factors, reducing cell proliferation, and facilitating apoptosis in response to exogenous stimulation (PubMed:12686563).
Related references: PMID:12686563 - Localizes to the endoplasmic reticulum, Golgi apparatus, Golgi stack, and is also found on the cell surface and secreted into the extracellular space.
- Shows highest expression in testis, stomach, skeletal muscle, lung, kidney, pancreas, small intestine, and colon. Down-regulation is observed in ovarian carcinoma, breast, pancreatic, renal, and hepatocellular carcinoma cell lines, indicating a potential tumor suppressor role.
- Key biological processes include apoptosis, calcium binding, and metal-binding; the protein is glycosylated and contains a signal peptide for secretion.
- Molecular weight of the full-length protein is approximately 101 kDa, consistent with the predicted 101027 Da from sequence.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 480–610 within the C-terminal region of human SULF1, which may provide specific detection of the target protein.
- For Western blot analysis, the predicted molecular weight is approximately 101 kDa; a band near this size is expected. Consider using appropriate positive controls, such as lysates from tissues with high SULF1 expression (e.g., testis, stomach).
- This antibody is suitable for ELISA applications; we recommend performing a checkerboard titration to determine optimal coating and detection antibody concentrations.
- Cross-reactivity is observed with rat SULF1, making it suitable for studies in rat models; validate in your specific sample system before routine use.
- The product is shipped on ice bags to maintain stability; store at the recommended temperature upon arrival.
