Biological Background: SUFU Function and Localization
- SUFU (Suppressor of Fused Homolog), also known as SUFUH, SUFUXL, PRO1280, and JBTS32, is a key component of the Hedgehog signaling pathway. Multiple isoforms are produced by alternative splicing.

- Classified as a tumor suppressor protein; post-translational modifications include phosphorylation, acetylation, and ubiquitination. Mutations are associated with ciliopathies such as Joubert syndrome 32 (JBTS32).
- May also function as a negative regulator of beta-catenin signaling, contributing to developmental processes and epidermal differentiation.
Experimental Guidance and Technical Tips
- Based on the immunogen design (C-terminal region, amino acids 325–484), the antibody is expected to recognize both major isoforms of SUFU. For Western blot, a predominant band around 54 kDa should be observed; minor bands representing isoforms or post-translationally modified forms may also be detected.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), consider antigen retrieval conditions and appropriate fixation methods. Since SUFU localizes to cytoplasm, nucleus, and cilia, ensure your imaging protocol can resolve these compartments.
- The antibody has been validated on human, mouse, and rat samples. For ELISA applications, use recombinant SUFU protein or cell lysates as coating antigens, and confirm specificity with appropriate controls.
