Biological Background: STYX Function and Localization
- Catalytically inactive phosphatase, also known as inactive tyrosine-protein phosphatase STYX or phosphoserine/threonine/tyrosine interaction protein.
- Acts as a nuclear anchor for MAPK1/MAPK3 (ERK1/ERK2), modulating cell-fate decisions and cell migration through spatiotemporal regulation of ERK signaling. Related references: PMID:23847209
- Binds to the F-box of FBXW7, preventing assembly of the SCF E3 ubiquitin-protein ligase complex and thereby inhibiting degradation of FBXW7 substrates. Related references: PMID:28007894
- Plays a role in spermatogenesis (by similarity).
- Predominantly localized in the nucleus, but also detected in the cytoplasm and cytosol.
- Identified as a phosphoprotein in proteomics studies; three-dimensional structure has been determined.
- Belongs to the protein tyrosine phosphatase family but lacks catalytic activity due to active site substitutions.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal 223 amino acids of human STYX; reactivity against endogenous protein should be validated in the relevant sample system.
- For Western blotting, a band at approximately 25 kDa is expected; consider using appropriate loading controls and optimizing blocking conditions.
- In ELISA applications, cross-reactivity with other phosphatases may be minimal but should be assessed if multi-target detection is performed.
- Optimal working dilutions for WB and ELISA must be determined empirically by the end user.
