Biological Background: Syntaxin-binding Protein 3 Function and Localization
- Syntaxin-binding protein 3 (STXBP3, also known as MUNC18C, PSP, Unc-18C) is a member of the Sec1/Munc18 protein family that regulates SNARE-mediated vesicle docking and fusion.
- STXBP3 together with STX4 and VAMP2 may play a role in insulin-dependent GLUT4 trafficking—mediating movement and docking/fusion of GLUT4-containing vesicles at the adipocyte cell surface.
- Expressed primarily in megakaryocytes and platelets; participates in platelet exocytosis.
- Subcellularly localizes to the cytoplasm and cytosol, and can associate with the cell membrane upon stimulation.
- Key functional domains and post-translational modifications, including phosphorylation, modulate its interaction with syntaxin partners and vesicle fusion.
- Relevant keywords: Cell membrane, Cytoplasm, Membrane, Phosphoprotein, Protein transport, Transport.
Experimental Guidance and Technical Tips
- The immunogen spans residues 343–592, a region that may be conserved across species, consistent with the predicted cross-reactivity to mouse and rat STXBP3.
- For western blotting, a band near 68 kDa is expected; consider using positive controls from human, mouse, or rat cell lysates, particularly adipocytes or platelet-rich samples.
- In ELISA applications, the antibody may detect native or recombinant STXBP3; validate in the relevant sample matrix and consider blocking peptide competition to confirm specificity.
- Non-reducing conditions or optimized sample preparation may improve detection depending on the target’s phosphorylation state and subcellular localization.
