Biological Background: STX17 Function and Localization
- STX17 (Syntaxin-17) is a member of the syntaxin family of SNARE proteins, which play essential roles in intracellular membrane fusion.
- Acts as a key SNARE on autophagosomes, directly mediating the fusion of autophagosomes with lysosomes to form autolysosomes, a critical step in autophagy. Related references: PMID:23217709, PMID:25686604, PMID:28306502
- May also function in the early secretory pathway, possibly contributing to the structural maintenance of the ER–Golgi intermediate compartment (ERGIC) and Golgi apparatus, and/or regulating trafficking between the endoplasmic reticulum, ERGIC, and Golgi. Related references: PMID:21545355
- Exhibits a complex subcellular distribution, localizing to membranes of the smooth endoplasmic reticulum, ERGIC, autophagosomes, COPII-coated vesicles, mitochondria, and autolysosomes, as well as in the cytosol.
- The human STX17 protein has a predicted molecular weight of approximately 33 kDa.
- Post-translational modifications including acetylation and phosphorylation have been identified, which may regulate its function or localization.
Experimental Guidance and Technical Tips
- The immunogen, corresponding to the N-terminal region (amino acids 1–130), is ideal for detecting total STX17 across applications.
- For Western blot, anticipate a band at ~33 kDa; optimize blocking and antibody dilution for your specific sample type and consider using a low-background detection system.
- In immunofluorescence/ICC, expect punctate staining patterns indicative of autophagic vesicles or ER/Golgi compartments; counterstaining with organelle markers can aid interpretation.
- Validate antibody performance in your experimental system, preferably using STX17 knockout or knockdown controls to confirm specificity.
- ELISA application may be useful for quantifying STX17 levels in cell lysates or tissue homogenates; ensure proper antigen coating and detection conditions.
