Biological Background: STRA13/CENPX Function and Localization
- Encoded by the CENPX gene, STRA13 (also known as CENP-X, MHF2, FAAP10, D9) is a histone-fold protein that functions as a DNA-binding component of the Fanconi anemia (FA) core complex and as a subunit of the constitutive centromere-associated network PMID:20347428.
- In the FA pathway, the STRA13-containing MHF heterodimer (with CENPS) is crucial for FANCM-mediated DNA remodeling, promotes gene conversion at blocked replication forks, and is required for FANCI-FANCD2 monoubiquitination and cellular resistance to DNA cross-linking agents PMID:20347429.
- As a component of the CENP-T-W-S-X heterotetramer, STRA13 contributes to kinetochore outer plate assembly and is essential for stable kinetochore-microtubule attachments and faithful chromosome segregation during mitosis PMID:19620631.
- STRA13 binds DNA with a preference for Holliday junctions and branched structures, but only in complex with its partner CENPS; it lacks intrinsic DNA-binding activity on its own PMID:20347429.
- Localized to the nucleus and concentrated at centromeres and kinetochores, STRA13 operates at the interface of DNA repair and chromosome segregation, linking genome maintenance with cell division.
- The protein is subject to acetylation and exists in alternatively spliced isoforms, with a calculated mass of approximately 9 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–63) of human STRA13; this sequence is identical to the full-length protein, suggesting the antibody recognizes the native STRA13 and possibly its isoforms.
- For western blotting, consider using whole-cell lysates from human cell lines and note that endogenous STRA13 migrates around 9 kDa; optimize blocking conditions and antibody dilution experimentally.
- This antibody is validated for ELISA; pairing with a suitable detection antibody may allow development of sandwich immunoassays for STRA13 quantification in biological fluids.
- Cross-reactivity has been tested against human samples; if you plan to use it in other species, validate specificity in the relevant system.
