Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide corresponding to the N-terminal region (amino acids 1-100) of human STK3/MST2. When using this antibody on mouse samples, validate specificity as cross-reactivity is confirmed but sequence homology may vary slightly.
- This antibody is suitable for Western blot (WB), immunofluorescence/immunocytochemistry (IF/ICC), and ELISA. For WB, consider using whole-cell lysates from human or mouse cell lines known to express STK3/MST2, and ensure proper blocking and antibody dilution optimization.
- For IF/ICC, fixation and permeabilization conditions should be optimized to detect nuclear and cytoplasmic pools of STK3/MST2. As STK3/MST2 can translocate to the nucleus upon caspase cleavage, monitoring apoptotic stimuli may enhance nuclear signal.
- Given its role in centrosome disjunction, this antibody may also be useful for centrosome studies; consider appropriate cell synchronization protocols and high-resolution microscopy.
