Biological Background: STK39 Function and Localization
- STK39 (STE20/SPS1-related proline-alanine-rich protein kinase) is an effector serine/threonine-protein kinase component of the WNK-SPAK/OSR1 kinase cascade involved in ion transport, response to hypertonic stress, and blood pressure regulation. Related references: PMID:16669787, PMID:18270262, PMID:21321328
- It specifically recognizes and binds proteins with an RFXV motif and acts downstream of WNK kinases (WNK1-4) to phosphorylate ion cotransporters such as SLC12A1/NKCC2, SLC12A2/NKCC1, SLC12A3/NCC, SLC12A5/KCC2, and SLC12A6/KCC3, thereby regulating their activity. Related references: PMID:16669787, PMID:21321328
- Mediates regulatory volume increase in response to hyperosmotic stress by catalyzing phosphorylation of ion cotransporters SLC12A1/NKCC2, SLC12A2/NKCC1, and SLC12A6/KCC3 downstream of WNK1 and WNK3. Related references: PMID:12740379, PMID:16669787, PMID:21321328
- Phosphorylates Na-K-Cl cotransporters SLC12A1 and SLC12A2 to promote activation and ion influx, while simultaneously inhibiting K-Cl cotransporters SLC12A5 and SLC12A6 to block ion efflux. Related references: PMID:16669787, PMID:19665974, PMID:21321328
- Acts as a key regulator of NaCl reabsorption in the distal nephron by mediating phosphorylation and activation of the thiazide-sensitive Na-Cl cotransporter SLC12A3/NCC downstream of WNK4. Related references: PMID:18270262
- Subcellular localization is predominantly cytoplasmic and nuclear.
- Predominantly expressed in brain and pancreas, with lower levels in heart, lung, kidney, skeletal muscle, liver, placenta, and testis.
- The protein undergoes acetylation and alternative splicing, and is classified as an ATP-binding phosphoprotein kinase.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide from amino acids 500–600 of human STK39, targeting the C-terminal region. Peptide competition assays may be considered to confirm binding specificity.
- For Western blot, the predicted molecular weight is 59 kDa. Validate the antibody in your specific sample system, as post-translational modifications or alternative splice variants can produce additional bands.
- In ELISA, optimization of coating antigen concentration and detection conditions is recommended to achieve optimal signal-to-noise ratios.
- As a polyclonal antibody, it may recognize multiple epitopes, potentially offering robust detection in both denaturing and native conditions. Assess lot-to-lot consistency for critical applications.
