Biological Background: STK39 Function and Localization
- STK39 (Serine/threonine-protein kinase 39), also known as SPAK or DCHT, is a 59 kDa effector kinase of the STE20 family with proline-alanine-rich regions.
- It functions as a key component of the WNK-SPAK/OSR1 kinase cascade, phosphorylating ion cotransporters such as SLC12A1/NKCC2, SLC12A2/NKCC1, SLC12A3/NCC, SLC12A5/KCC2, and SLC12A6/KCC3 to regulate their activities. Related references: PMID:16669787, PMID:21321328
- STK39 mediates regulatory volume increase by simultaneously activating Na-K-Cl cotransporters (NKCC1, NKCC2) and inhibiting K-Cl cotransporters (KCC2, KCC3) downstream of WNK1 and WNK3 kinases. Related references: PMID:16669787, PMID:19665974, PMID:21321328
