Biological Background: STK38 Function and Localization
- STK38 (Serine/threonine-protein kinase 38), also known as NDR1 protein kinase, is a ubiquitously expressed kinase with highest levels in peripheral blood leukocytes.
- It acts as a negative regulator of MAP3K1/2 signaling by converting MAP3K2 to its non-phosphorylated form and inhibiting autophosphorylation. Related references: PMID:12493777 PMID:15197186 PMID:17906693
- In response to DNA damage, STK38 functions as a ufmylation reader, binding mono-ufmylated histone H4 and recruiting SUV39H1 to double-strand breaks, leading to ATM activation. Related references: PMID:32537488
- Subcellularly, STK38 localizes to the nucleus, cytoplasm, and chromosomes, consistent with its roles in signaling and DNA damage repair.
- The protein contains kinase, ATP-binding, and metal-binding domains, and undergoes acetylation and phosphorylation, as indicated by proteomics and direct protein sequencing.
- STK38 is involved in Ubl conjugation pathways and may have broader regulatory functions in cell cycle and metabolism, given its kinase activity and DNA damage association.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a region within the C-terminal domain (aa 366–465); consider this when analyzing truncated or mutant forms.
- For WB, a band at ~54 kDa is expected; validate in the relevant sample system.
- For IF/ICC, ensure appropriate permeabilization to access nuclear and cytoplasmic targets.
- Cross-reactivity with mouse and rat is reported; verify performance in your model.
- Optimal dilutions and incubation times should be determined empirically for each application.
