Biological Background: Serine/threonine-protein kinase 36 (STK36) Function and Localization

  • STK36 (serine/threonine-protein kinase 36), also known as Fused homolog, is a protein kinase encoded by the STK36 gene. Alternative splicing produces multiple isoforms.
  • Plays a critical role in the Hedgehog signaling pathway by regulating GLI transcription factors. STK36 opposes the negative regulator SUFU and promotes nuclear localization of GLI1, GLI2, and GLI3. Related references: PMID:10806483, PMID:31279575, PMID:38096226
  • STK36 directly phosphorylates GLI proteins, facilitating their dissociation from SUFU, a key step for transcriptional activation. Related references: PMID:28543983, PMID:31279575, PMID:38096226
  • GLI2 requires an additional function of STK36 beyond its kinase activity to become transcriptionally active. Related references: PMID:10806483
  • Localizes to the cilium, axonemal cytoskeleton, cytoplasm, and nucleus, consistent with its roles in signaling and ciliary function.
  • Highly expressed in adult testis germ cells and respiratory epithelial cells; low expression in most fetal tissues and adult ovaries. Related references: PMID:10806483, PMID:28543983
  • Essential for postnatal development, construction of the central pair apparatus of motile cilia, and implicated in primary ciliary dyskinesia (CILD46). Related references: PMID:28543983

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 250-450 of human STK36 (NP_056505.2); the antibody may detect multiple isoforms due to alternative splicing, so validate in your sample system.
  • For Western blot, a band around 144 kDa is expected; consider that post-translational modifications or isoforms may produce additional bands.
  • Cross-reactivity with mouse is indicated, making this antibody suitable for murine model studies; verify performance under your experimental conditions.
  • Optimize antibody dilution for WB and ELISA; recommended starting dilutions should be determined empirically.
