Biological Background: STK19 Function and Localization
- STK19 (WHR1), also known as Protein G11, Protein RP1, and formerly as serine/threonine-protein kinase 19, is now classified as an inactive kinase and a winged helix repair factor.
- Acts as a DNA-binding protein essential for transcription-coupled nucleotide excision repair (TC-NER), functioning within a complex that assembles on RNA polymerase II (RNAPII) stalled at DNA lesions.
Related references: PMID:38252411, PMID:38890355, PMID:39353615 - Facilitates recruitment of TFIIH to TC-NER complexes by interacting with the XPD helicase subunit, guiding TFIIH to downstream DNA to coordinate repair.
Related references: PMID:39353615, PMID:39547228, PMID:39547229 - Directly binds RNAPII and downstream DNA, promoting timely removal of damage-stalled RNAPII to allow NER factor access to lesions.
Related references: PMID:39547228, PMID:39547229 - Required for UVSSA monoubiquitination and stimulates RBP1 (RNAPII subunit) ubiquitination, regulating TC-NER complex stability and RNAPII processing.
Related references: PMID:39353615, PMID:39547223 - Also exhibits RNA-binding activity and modulates the expression of numerous mRNAs.
Related references: PMID:38252411, PMID:38890355 - Localizes predominantly to the nucleus but is also detected in the cytoplasm, and is expressed in monocytes, hepatocytes, epithelial cells, and T- and B-lymphocytes.
Experimental Guidance and Technical Tips
- The immunogen (aa 75-364) targets a central conserved region, supporting cross-reactivity with mouse (GeneID: 192641) and rat (GeneID: 296356) as indicated; confirm sequence homology in the target species.
- For Western blot (WB), a band at 41 kDa is expected; include proper positive controls such as HeLa or Jurkat cell lysates.
- In immunohistochemistry (IHC-P), consider testing on liver, spleen, or thymus sections where STK19 is enriched; validate with appropriate isotype controls.
- For ELISA, titrate the antibody to determine the optimal working concentration for your specific plate coating and detection system.
