Biological Background: STIP1 Function and Localization
- Stress-induced-phosphoprotein 1 (STIP1), also known as Hsc70/Hsp90-organizing protein (Hop), renal carcinoma antigen NY-REN-11, or transformation-sensitive protein IEF SSP 3521, is a 63 kDa co-chaperone encoded by the STIP1 gene.
- Acts as a co-chaperone for HSP90AA1 and mediates the association of the molecular chaperones HSPA8/HSC70 and HSP90. Related references: PMID:27353360
- The protein shuttles between the cytoplasm and nucleus, and is also found in dynein axonemal particles, reflecting its dynamic role in chaperone complexes.
- STIP1 contains multiple tetratricopeptide repeat (TPR) domains that facilitate protein–protein interactions with heat shock proteins.
- Post-translational modifications include phosphorylation and acetylation, which may regulate its co-chaperone activity and subcellular distribution.
- Referenced in proteomics identification and alternative splicing databases, with evidence at protein level, supporting its broad physiological relevance.
Experimental Guidance and Technical Tips
- The immunogen was designed against amino acids 1–300 of human STIP1; consider this region when assessing epitope accessibility in fixed or native conformations.
- For WB, a band around 63 kDa is expected in lysates from human, mouse, rat, or monkey cells; verify using appropriate positive controls.
- IHC-P and IF/ICC may require antigen retrieval or permeabilization optimization; validate staining patterns against known subcellular localizations (cytoplasmic/nuclear).
- In IP experiments, include appropriate IgG controls and consider the co-chaperone complex dynamics; co-precipitation of HSP90 or HSC70 may be observed.
