Biological Background: STING/TMEM173 Function and Localization
- STING (Stimulator of interferon genes protein), also known as TMEM173, ERIS, MITA, MPYS, is encoded by the STING1 gene. It acts as a pivotal cytosolic DNA sensor in innate immunity.
- Functions as a facilitator of innate immune signaling by sensing non-CpG double-stranded DNA from viruses and bacteria, promoting production of type I interferons (IFN-α and IFN-β). Related references: PMID:18724357 PMID:18818105 PMID:19433799
- Recognizes and binds cyclic dinucleotides, including bacterial c-di-GMP and endogenous 2',3'-cGAMP produced by cGAS in response to cytosolic DNA. STING preferentially recognizes 2',3'-linked cGAMP due to its organized conformation that pre-matches the binding pocket. - Upon ligand binding, STING oligomerizes, translocates from the endoplasmic reticulum, and is phosphorylated by TBK1 on the pLxIS motif, leading to recruitment and activation of IRF3 to induce type I interferons, establishing an antiviral state. - In addition to interferon induction, STING directly triggers autophagy. Following activation, it buds from the ER in COPII vesicles to form the ER-Golgi intermediate compartment (ERGIC), which serves as the membrane source for LC3 lipidation and autophagosome formation, targeting cytosolic DNA for lysosomal degradation. - Subcellular localization spans the endoplasmic reticulum, ERGIC, Golgi apparatus, autophagosome, mitochondrial outer membrane, and plasma membrane. STING also localizes to the perinuclear region and cytoplasmic vesicles.
