Biological Background: STAT4 Function and Localization
- STAT4 is a transcriptional regulator primarily expressed in hematopoietic cells, playing a critical role in cellular growth, differentiation, and immune responses. Related references: PMID:10961885, PMID:37256972, PMID:8943379
- It is essential for T-helper 1 (Th1) cell differentiation and the production of interferon-gamma (IFN-γ). Related references: PMID:12213961, PMID:35614130
- STAT4 participates in multiple neutrophil functions, including chemotaxis and the formation of neutrophil extracellular traps.
- Upon interleukin-12 (IL12) binding to its receptor IL12RB2, STAT4 interacts with the intracellular domain, becomes tyrosine-phosphorylated, homodimerizes, and translocates to the nucleus to regulate IL12-responsive genes. Related references: PMID:10415122, PMID:7638186
- STAT4 can also be phosphorylated and activated by IFN-gamma via JAK1 and TYK2, and by interleukins IL23, IL2, and IL35. Related references: PMID:11114383, PMID:34508746
- Transcriptional activation of the IFN-gamma gene is mediated by interaction with JUN, forming a complex at the AP-1-related promoter sequence.
- In response to IFN-alpha/beta signaling, STAT4 acts as a transcriptional repressor, suppressing IL5 and IL13 mRNA during T-cell receptor (TCR) activation. Related references: PMID:26990433
- STAT4 localizes to both the cytoplasm and nucleus, and its activity is regulated by post-translational modifications including acetylation and phosphorylation; it contains an SH2 domain and is associated with systemic lupus erythematosus.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a synthetic peptide spanning amino acids 649–748 of human STAT4 (NP_003142.1), located in the C-terminal region. This design is suitable for detecting full-length STAT4 protein.
- For Western blotting, a band at approximately 86 kDa is expected. Verify specificity by using appropriate positive and negative controls.
- For immunofluorescence/immunocytochemistry, optimize fixation and permeabilization conditions for your cell type to ensure proper antigen accessibility.
- For ELISA applications, consider pairing with a suitable detection antibody and validate performance with recombinant STAT4 or cell lysates.
- Cross-reactivity with mouse and rat STAT4 has been observed, but sequence alignment is recommended to confirm epitope conservation in your species of interest.
