Biological Background: STAT3 Function and Localization
- STAT3 is a signal transducer and transcription activator that mediates cellular responses to interleukins and growth factors, playing a central role in the JAK/STAT signaling pathway. Related references: PMID:10688651, PMID:11294841, PMID:12359225
- It activates transcription by recruiting coactivators like NCOA1 or MED1 to target gene promoters. Related references: PMID:15653507, PMID:16285960, PMID:17344214
- STAT3 is essential for inflammatory responses, regulating differentiation of naive CD4+ T-cells into Th17 or Treg cells. Related references: PMID:28065600, PMID:28262505
- It is involved in cell cycle regulation by inducing expression of G1/S phase transition genes such as CCND1. Related references: PMID:17344214
- STAT3 shuttles between the cytoplasm and nucleus (Subcellular location: Cytoplasm, Nucleus).
- Expressed widely in tissues including heart, brain, placenta, lung, liver, skeletal muscle, kidney, and pancreas, as well as in naive and differentiated T-cells.
- Post-translational modifications include acetylation, methylation, phosphorylation, and ubiquitination, as reflected by keywords: Acetylation, Methylation, Phosphoprotein, Ubl conjugation.
- Plays a role in apoptosis and autophagy regulation. Related references: PMID:18242580, PMID:23084476
Experimental Guidance and Technical Tips
- The immunogen used is a synthetic peptide spanning amino acids 611-710 of human STAT3 (NP_003141.2), located in the C-terminal region. This region may be conserved across isoforms; verify reactivity with relevant controls.
- For western blot (WB), a band near 88kDa is expected. Use appropriate loading controls and consider phosphatase inhibitors if assessing phospho-STAT3 status.
- In immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), STAT3 can display both cytoplasmic and nuclear localization. Optimize fixation and permeabilization conditions to preserve antigenicity.
- The antibody is reported to cross-react with human, mouse, rat, and monkey STAT3, making it suitable for multi-species studies. Validate specificity in the species of interest.
- For ELISA applications, ensure the antibody is compatible with the detection format (e.g., direct, indirect, or sandwich ELISA) and titration is performed.
