Biological Background: STAT3 Function and Localization

Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal peptide (aa 600-700) of human STAT3. Consider that this region may be subject to post-translational modifications that could affect antibody binding; use appropriate phosphatase or deacetylase inhibitors if necessary.
- For Western blot, use standard sample preparation and include a phosphatase inhibitor cocktail to preserve phosphorylation states. A primary antibody dilution in the range of 1:500-1:2000 is commonly used for similar antibodies, but individual optimization is recommended.
- For ELISA, validate the antibody with recombinant STAT3 protein or the immunizing peptide to ensure specificity. The antibody may bind both human and mouse STAT3; confirm cross-reactivity in your assay system.
- Note that the predicted molecular weight is 88 kDa; however, due to phosphorylation and other modifications, the observed band may shift or appear as multiple bands. Include appropriate controls.
