Biological Background: STAMBP Function and Localization
- Protein identity: STAMBP, also known as AMSH (Associated molecule with the SH3 domain of STAM) and MICCAP, is an endosome-associated ubiquitin isopeptidase encoded by the STAMBP gene.
- Lys-63-specific deubiquitinase: It is a zinc metalloprotease that specifically hydrolyzes 'Lys-63'-linked polyubiquitin chains, with no activity toward 'Lys-48' linkages (PubMed:15314065, PubMed:23542699, PubMed:34425109).

- For Western blotting, the predicted molecular weight is approximately 48 kDa. Consider the potential impact of phosphorylation or other post-translational modifications on electrophoretic mobility, and use appropriate positive and negative controls.
- When performing immunohistochemistry or immunocytochemistry, antigen retrieval conditions and blocking steps should be optimized. Given STAMBP’s localization to multiple compartments, include subcellular markers in your analysis.
- In immunoprecipitation assays, maintain native protein–protein interactions; STAMBP interacts with components of the endosomal sorting machinery, which may require mild lysis conditions.
- This polyclonal antibody may be used in ELISA; however, direct detection limits should be determined empirically for the target antigen and assay format.
