Biological Background: ST8SIA4 Function and Localization

  • ST8SIA4 (also known as PST, PST1, SIAT8D, or ST8Sia IV) is a polysialyltransferase that catalyzes the transfer of sialic acid from CMP-sialic acid to terminal alpha-2,3-, alpha-2,6-, or alpha-2,8-linked sialic acid residues on N-glycans, forming alpha-2,8-linked polysialic acid chains on glycoproteins such as NCAM1, FETUB, and AHSG. Alpha-2,3-linked acceptors are preferred over alpha-2,6-linked substrates. Related references: PMID:10766765, PMID:11279095, PMID:28810663, PMID:9774483
  • ST8SIA4 belongs to the sialyltransferase 8 (ST8Sia) family and is the fourth member (ST8Sia IV), essential for polysialic acid synthesis in mammals.
  • The protein localizes primarily to the Golgi apparatus membrane via a signal-anchor transmembrane domain, but a soluble, secreted form also exists.
  • It contains a disulfide bond and is N-glycosylated, which may influence its activity and stability.
  • ST8SIA4 is highly expressed in fetal brain, lung, and kidney, and in adult heart, spleen, and thymus, with lower levels in adult brain, placenta, lung, intestine, and peripheral blood leukocytes. Related references: PMID:7624364
  • Alternative splicing generates multiple isoforms, providing functional diversity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 20-168 of human ST8SIA4; this region may be suitable for detecting the protein in Western blot (WB) and ELISA applications.
  • A band at approximately 41 kDa is expected in WB; consider using appropriate molecular weight markers and validating the signal in ST8SIA4-expressing cell lines or tissues.
  • For ELISA, titration of both coating antigen and antibody concentration is recommended to optimize the signal-to-noise ratio.
  • As with polyclonal antibodies, lot-to-lot variability should be assessed, and appropriate negative controls (e.g., knockout cell lysates) should be included to confirm specificity.
