Biological Background: ST6GAL1/CD75 Function and Localization

  • ST6GAL1, also known as CD75, ST6N, SIAT1, and ST6GalI, is a beta-galactoside alpha-2,6-sialyltransferase that catalyzes the transfer of sialic acid to galactose-containing acceptor substrates.
  • In B lymphocytes, this enzyme generates neuraminidase-sensitive cell-surface differentiation antigens, including CDw75, HB-6, and CD76 (PubMed:1730763). Related references: PMID:1730763
  • ST6GAL1 sialylates complex-type N-glycans on the Fc region of immunoglobulins, conferring anti-inflammatory effector functions to IgG.
  • The enzyme preferentially monosialylates the alpha(1->3) mannose antenna of Fc glycoforms, with subsequent disialylation occurring at a much slower rate.
  • Subcellularly, ST6GAL1 localizes to the Golgi stack membrane, consistent with its role in glycan processing, and can also be secreted.
  • Alternative splicing produces multiple isoforms, with this antibody recognizing forms at approximately 20 kDa and 47 kDa, as indicated by the product data.
  • Post-translational modifications reported for ST6GAL1 include glycosylation, disulfide bond formation, and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 44-406 of human ST6GAL1, covering nearly the entire extracellular domain; this broad region may allow detection of multiple isoforms.
  • For Western blotting, consider the predicted molecular weights (20 kDa and 47 kDa) and validate detection in your sample system, optimizing protein loading and antibody concentration as needed.
  • In immunohistochemistry (IHC-P) and immunocytochemistry (IF/ICC), ensure appropriate fixation and permeabilization protocols to preserve antigenicity.
  • Flow cytometry (FC) and ELISA applications suggest the antibody recognizes native protein; include proper isotype controls and validation with known positive and negative samples.
